Mayor Michael Bloomberg says the streets will be closed to vehicles from 7 a.m. to 1 p.m. on three Saturdays in August.
Several crosstown streets will still be open.
The program is modeled after recreation areas in other cities around the world - including Bogota, Colombia; Guadalajara, Mexico; and El Paso, Texas.
The route will include rest stops offering water and bike repairs, as well as activities like fitness, dance and yoga.
