Authorities say the 70-year-old driver suffered a medical episode, he lost control of the vehicle and flipped over a barrier at the intersection and landed in the trainyard below.
The driver was pronounced dead at the scene while the female passenger was taken to Brooklyn Methodist Hospital in critical condition.
The crash scene is a pedestrian-heavy area with shops and restaurants near the Barclays Center.
Fire officials say they believe the car fell about 30 feet.
The area is supposed to be covered as part of the massive Pacific Park development project which will be a mix of condos and affordable housing.
The project has been has been delayed several times and members of the community say the deadly accident could have been avoided.
"I feel like whoever owns this, they should find a way to make it safer because anyone could climb over this gate and jump, anyone can hit the brick and fall over," resident Kimberly Damus said.
