It happened on the Southern State Parkway near Exit 29 in North Massapequa shortly after 11 p.m.
Police say a black 2016 Dodge Dart was speeding and traveling in and out of lanes of traffic before it lost control and crashed onto a tree on the shoulder.
The driver, 19-year-old Jaden Dsouza, and front-seat passenger 23-year-old Anthonie Marte, were extracted from the vehicle and taken to a hospital in serious condition.
Rear passengers Hailey Dsouza, 21, and Crystal Alba-Figueroa, 23, were pronounced dead at the scene.
Dsouza was charged with driving while ability impaired by drugs, manslaughter, vehicular manslaughter, assault and aggravated vehicular homicide.
Eastbound lanes were closed for more than six hours, but reopened for the morning commute.
