It happened at 996 Amsterdam Ave. between West 109th Street and Cathedral Avenue around 6 p.m.
Police say a vehicle traveling northbound on Amsterdam struck some parked cars, including a gray Volkswagen and brown minivan, before striking five men.
They say the vehicle was a driven by a man who lost control of his car.
Two of the five men were killed, and the conditions of the other three are unclear.
Everyone, including the driver, was taken to St. Luke's Hospital.
Individuals inside the parked minivan, possibly two men, refused medical attention.
Police say they do not believe the crash was intentional.
No arrests have been made so far.
The investigation is ongoing.
