67-year-old Janice Ross was on oxygen and overcome by smoke after flames broke out just after 2 a.m. Monday, officials said.
An 8-year-old girl was also injured in the blaze at a building on East 92nd Street.
She was taken to a local hospital to be treated for a minor injury.
Investigators are still working to determine what sparked the fire.
It broke out around the same time as an explosive fire that engulfed a parking garage in East New York.
The parking garage fire damaged up to eight cars.
