The fire happened at a building on Vermont Street in the East New York section at 1:15 a.m.
The flames started in the first-floor tire shop and then spread to the apartment on the second floor, investigators said. The cause is not known.
Two residents became trapped by the flames.
One man was pronounced dead at the hospital, while another was listed in critical condition.
A cat survived the fire after receiving oxygen from firefighters, officials said.
